Crassocephalum rabens (Asteraceae) is a common herb used in Taiwanese folk medicine to treat inflammation-related syndromes. Pharmacological studies have revealed that galactolipids exhibit anti-oxidative, anti-inflammatory, and anti-hyaluronidase activities and improve skin wrinkles, moisture, and elasticity in healthy subjects. However, the anti-aging effects of C. rabens and its primary active compound, 1,2-di-O-linolenoyl-3-O-β-galactopyranosyl-sn-glycerol (dLGG), remain elusive. Here, we investigated whether C. rabens can improve skin conditions in healthy individuals using a double-blind approach. Forty enrolled volunteers were randomly and equally assigned to the control or treatment group and were required to take either a placebo or a C. rabens extract capsule daily for one month. Skin parameters were measured before and after the study. The results showed significant differences in skin elasticity, wrinkles, collagen content, brightness, and hydration between the baseline and week 4 in the treatment group. Particularly, compared with those in the placebo group, skin wrinkles (p < 0.05), brightness (p < 0.001), collagen content (p < 0.01), and UV spots (p < 0.05) were notably improved after treatment with the C. rabens extract. Our study successfully demonstrated the application of C. rabens in preventing skin aging. Further investigations will be conducted to study the underlying anti-aging mechanism of dLGG....
Skin aging usually results from intrinsic or extrinsic stress. Photodamage promotes skin damage and stimulates the skin, manifesting as wrinkles, dryness, roughness, and loss of elasticity. We have previously found that blackberry (Rubus fruticosus B) fermented by Lactobacillus plantarum JBMI F5, designated BB-1000, showed an in vitro and in vivo anti-skin-aging activity. In the present study, we have further evaluated the anti-aging effect of BB-1000 via a randomized, double-blind, and placebo-controlled clinical trial. The trial included 102 volunteers aged 35 to 59 years who have dry skin and wrinkles. Subjects took BB-1000 or a placebo orally at 800 mg/day for 12 weeks. Skin hydration and degree of wrinkles around the eyes were measured at weeks 6 and 12. Skin hydration had no significant effect in both groups at weeks 6 and 12. Otherwise, volunteers in the BB-1000 group had a significant reduction in eye wrinkle grade at week 12. These findings suggest that BB-1000 may be considered a candidate anti-aging agent for preventing skin wrinkles as a nutricosmetic agent....
Skin disorders are widespread around the world, affecting people of all ages, and oxidative stress represents one of the main causes of alteration in the normal physiological parameters of skin cells. In this work, we combined a natural protein, fibroin, with antioxidant compounds extracted in water from pomegranate waste. We demonstrate the effective and facile fabrication of bioactive and eco-sustainable films of potential interest for skin repair. The blended films are visually transparent (around 90%); flexible; stable in physiological conditions and in the presence of trypsin for 12 days; able to release the bioactive compounds in a controlled manner; based on Fickian diffusion; and biocompatible towards the main skin cells, keratinocytes and fibroblasts. Furthermore, reactive oxygen species (ROS) production tests demonstrated the high capacity of our films to reduce the oxidative stress induced in cells, which is responsible for various skin diseases....
Plant extracts have long served as important sources of bioactive compounds, and they are currently the focus of extensive research in the development of novel preventive and therapeutic strategies. However, their health benefits are often limited by low bioavailability. Nanoparticle delivery systems can represent a solution to such limitations. Euphorbia characias is a Mediterranean shrub known to have biological activities, such as inhibiting tyrosinase and showing a potential role as a skin-whitening agent. In this study, an ethanolic extract from E. characias leaves was tested for its inhibitory activity on skin-related enzymes, such as elastase, collagenase, and hyaluronidase, and for sun protection factors. Moreover, the extract was formulated in phospholipid vesicles to improve its local bioavailability and applicability. The vesicles were characterized by size, surface charge, storage stability, and entrapment efficiency. The nanoformulation was also evaluated for antioxidant activity and assayed for cytocompatibility and anti-tyrosinase activity in melanoma cells. Our findings demonstrated that the extract has a photo-protective effect and enzyme-inhibitory properties. E. characias nanoformulation was also cytocompatible and improved the extract’s activity in the cells, suggesting a potential skin application for antimelanogenic treatments and confirming the key role of nanotechnological approaches to maximize plant extract’s potentialities....
Glycosides have been synthesized using the starting materials magnolol (1) and honokiol (4), isolated from the Japanese white-bark magnolia, and their anti-aging effects on the skin (skinbeautifying effects) have been examined. The advanced glycation end-product (AGE) inhibitory activity test (anti-glycation test) and glycation induction model test, using human-derived dermal fibroblasts, TIG-110 cells, have been conducted to evaluate the anti-aging effects. The synthesized glycoside compounds, 5,5-di(prop-2-en-1-yl)[1,1-biphenyl]-2-hydroxy-2-glucopyranoside (3a), 5,5-di(prop-2- en-1-yl)[1,1-biphenyl]-2,2-diglucopyranoside (3b), 3,5-di(prop-2-en-1-yl)[1,1-biphenyl]-4-hydroxy-2- glucopyranoside (6a) and 3,5-di(prop-2-en-1-yl)[1,1-biphenyl]-2,4-diglucopyranoside (6b), have shown significant anti-glycation activities of less than 0.10 mM in IC50. The glycation induction model test with the fibroblasts, TIG-110 cells, demonstrates that the aforementioned glycosides significantly inhibit the decrease in cell viability.
